Estimating Island Fox Age Using Cementum Rings In Teeth

In October 2019, Friends of the Island Fox funded a research study by Stacy Baker and Juliann Schamel, via a donation from Safari West, to investigate the use of cementum analysis as a method to determine island fox age at death. The results are in and the answer is, well mostly yes, but...

To conduct the analysis 31 teeth from island foxes, whose age was known when they died, were sent off for testing. Of the 31 samples, 28 were aged accurately to within 1 year of their known age, two teeth were underestimated by 3 years and one was underestimated by 4 years. This underestimation primarily occurred with older foxes.

The data suggests that for younger and middle age foxes–up to approximately age 7–cementum analysis can provide an accurate assessment of age at the time of death. (A more accurate assessment than estimating age by tooth wear.) For more senior island foxes, however, the accuracy of the method drops significantly. 

What are the factors that led to a misread of the older fox teeth? 

According to Baker, the study's principal scientist: "Tooth condition may perhaps play a role, as two of those teeth out of four were determined to be of lower quality for aging according to the Matson's Lab. This also may be unavoidable, since broken and decayed teeth become more prevalent as individuals age." (Matson's Laboratory in Manhattan, Montana, specializes in aging mammals using teeth.) With the limited sample size from the older fox population, more analysis is warranted to tease out the factors that may play a role in the reading error. In the wild, foxes are known to live 10–12 years on some islands. (K Dearborn personal communication)

Island Foxes and Island Spotted Skunks Sharing Space On Santa Cruz Island

On the California Channel Islands, the island fox (Urocyon littoralis) has been hypothesized to compete with the smaller-bodied island spotted skunk (Spilogale gracilis amphiala). Recent declines in spotted skunk captures have led to concerns on population viability and what role the foxes may have played in the decline.

(Calypso worked in the field on Santa Cruz Island with fellow researcher Victor Zhang.

From 2018–2019, we GPS and VHF collared foxes and spotted skunks living on Santa Cruz Island to assess space use and deployed remote cameras to examine interactions between the two species at spotted skunk den sites. We also explored monitoring tools for spotted skunk detection with emphasis on remote camera placement and use of scent stations.

We found fox and spotted skunk seasonal home ranges were much larger than previously reported on Santa Cruz Island and spotted skunks moved around the landscape differently compared to foxes.  

Spotted skunks showed restricted movement, using less than 50% of their home range over shorter time periods such as a week or month. Foxed moved widely through the landscape covering more than 50% of their home range over the course of a week. During this time, we collected the first photo evidence that foxes may be disturbing resting sites of spotted skunks during the day. We also found that remote cameras placed on drainage bottoms may be more effective in detecting skunks. We discourage the use of scents at camera stations as foxes appear to be monopolizing the stations by repeated marking.

Our knowledge and understanding of spotted skunk ecology are still limited due to the many challenges of studying a cryptic species. We encourage further studies be conducted on spotted skunks, particularly focusing on interactions of foxes and spotted skunks at den sites.

Fox Foto Friday - Island Foxes in Washington D.C.?

What smells like an island fox? Came from an island fox? But is contained on the tip of a cotton swab? A sample of island fox microbiome.

Friends of the Island Fox recently heard from FIF 2020 Research Grant recipient, Alexandria DeCandia, Ph.D.

DeCandia wrote "I wanted to let you know that microbiome samples from all six islands have arrived on the east coast :)" Swabs taken from island foxes during health checks have been transferred to a lab in Washington D.C. She added, "I'm very much looking forward to the next steps of this research!"


FIF's Research Grant is funding the DNA analysis of the swabs to get a picture of the biodiversity of microbes living on and in island foxes.

What Is She Looking For?

DeCandia's research is cutting edge science. Her early analysis of Catalina Island foxes led to some important findings on the possible relationships between bacteria in island fox ear canals and a tendency toward extreme immune system response to ear mites. This next stage of research builds on that work.

Maintaining healthy island foxes across the islands requires greater in depth understanding of their health, diet, position in the ecosystem, and interactions with other species, including humans.

Gathering Specimens For Island Fox Tooth Research

In 2019 Friends of the Island Fox supported a research project investigating whether cementum ring structures in teeth could be used to accurately determine island fox age at death. Research Project by Stacy Baker and Juliann Schamel.

Baker and Schamel updated us on their progress mid-March 2020.

"We went up to the Santa Barbara Museum of Natural History last week to collect teeth from the first seven skulls that they are processing for their collection," Schamel wrote in mid-March. An additional five skulls have since been processed.


The researchers explained that the island fox skulls came from Dr. Leslie Woods and the CA Animal Health and Food Safety Lab at UC Davis, which is responsible for conducting necropsies on island foxes when they die on the Channel Islands. These specimens are vital for biologists to understand cause of death, as well as underlying health problems. When an island fox dies, a necropsy (similar to an autopsy for a human) examines organs and tissues in detail to understand the cause of death. Necropsies are vital to identifying disease or any other threat, which might endanger other island foxes. The bodies of these individual animals are stored in special freezers at UC Davis to preserve tissue samples for future scientific investigations, like the tooth project.

During the final stages of the preservation process, cleaned skulls are soaked in cold water for several days. The researchers explain that "At that point in the process, the teeth are very loose and easy to pull out..." A single lower canine tooth was extracted from each skull. Once all of the skulls have been processed, Schamel added, "I think we will be ready to send all the teeth to Matson's Laboratory in Montana!"

The plot below shows how fox age estimated by tooth wear does not always match-up with actual island fox age.


We know the actual age of island foxes that are micro-chipped as pups. Biologists performing health checks in the field, however, do not have access to this age information when they are examining individual foxes. They estimate the fox's age by the amount of wear on its teeth. This estimate puts the fox in one of five categories: 
  • Age Class 0 - pup to 1 year
  • Age Class 1 - young adult
  • Age Class 2 - adult
  • Age Class 3 - mature adult
  • Age Class 4 - senior
The plot above shows data from 1300 exams of island foxes compared with their known ages. The larger the dot the more individuals fell into each category. The more you examine this plot the more you will see. For example: a 4-year-old island fox was almost as likely to be estimated as Age Class 3 as Age Class 2. A few 4-year-olds were estimated as Age Class 1 or Age Class 4. What do you see in the plot for island foxes that are 6 years old? Is tooth wear a reliable way to estimate island fox age? 

This data shows why the cementum research is so important. Island foxes live longer and eat a different diet from the southeastern gray foxes for which the tooth-wear age classes were originally designed.


This project also highlights the broad scientific community involved in researching island foxes. Biologists from Channel Islands National Park are working with the Santa Barbara Natural History Museum, UC Davis, and Matson's Laboratory in Montana, which specializes in cementum age analysis of mammal teeth. Science brings people together.

As this research project moves forward FIF will keep you updated on the findings. Scientific data is vital to making informed conservation decisions that maintain sustainable populations of island foxes.

How Structures in Teeth Could Provide Accurate Age Information for Island Foxes?


In October 2019, Friends of the Island Fox funded a research study, via a donation from Safari West, to investigate the use of cementum analysis as a method to determine island fox age at death. The current standard method for estimating age in island foxes is based on the wear of the upper first molar. This method is imprecise as tooth wear is heavily influenced by diet and varies significantly between islands and even among habitats on the same island.

Knowing the age at death is very valuable as it allows the National Park and other island managers to track the island fox lifespan. This is important as a fox who dies at age 9 or 10, after reproducing for many years, has contributed to the survival of the species. A fox that dies at 3 or 4 may not have had a chance to replace itself in the population or pass on its genes.

Cementum is a thin mineralized tissue covering the root surface of teeth and functions as a tooth supporting device which anchors the tooth in the socket. Unlike bone, cementum is produced throughout one's life and forms annual layers (annuli)–a fact that makes it useful in aging techniques.

To determine the age of an island fox after it has died, the lower canine tooth is removed and sent to the lab for aging.

Once the tooth is received at the lab it is cleaned to remove any dirt and soft tissue, then undergoes processing to decalcify the tooth in preparation for analysis.

This makes the tooth very soft and pliable. The tooth is then preserved by fixing it in formaldehyde, also known as formalin, to preserve proteins and vital structures within the tooth. Next, it is embedded in a paraffin wax block which provides a support medium to make it easier to cut thin slices for examination.

The paraffin embedded tooth is cut into sections, approximately 15 microns thick (about the size of a droplet of mist or fog) on a device called a microtome, which is very similar to a deli slicer. The thin sections are mounted on glass microscope slides as the final step prior to analysis.

The sections are stained with a special histological dye that is taken up by annuli at varying degrees (light blue or darker blue) depending on the amount of cementum laid down in a given year. This provides both a highlight and contrast for the purposes of counting the number of growth rings (aging) under a microscope.

Significance
If it is possible to accurately determine a fox's age at time of death, it can be used to monitor average longevity and to investigate many questions across all six of the Channel Island. Questions such as:
  • What age group of foxes are most likely to be hit by cars?
  • How old were foxes collected during annual mortality monitoring?
  • Do males or females have a greater lifespan?

FIF Research Grant to Investigate Further Into Island Fox Diet




Friends of the Island Fox is happy to announce that Juliann Schamel's research investigating island fox diet through stable isotopes in whisker samples has been chosen to receive the FIF Research Grant for 2019.

This second year of support will enable analysis of additional data sets to identify seasonal dietary items during drought and important to successful reproduction. It has long been believed that deer mice are a vital food item during breeding and pupping season. Is it true? Does availability to a specific food source influence successful reproduction? If island foxes do not have accesses to abundant deer mice are they less successful as parents?

The grant will also expand the study to look at island fox use of marine resources. Do island foxes use marine foods during drought? If so, which island foxes are able to make use of marine resources?

Island fox whiskers provide an amazing record of what an individual island fox has been eating over 5–6 months. All food items are made up of carbon and nitrogen, but each kind of food has a different balance of elements and therefore a different signature. These individual isotope signatures are recorded in the fox's whisker. More on Schamel's initial research.

Schamel's initial whisker data set went to the lab this spring. At the Center for Stable Isotopes at the University of New Mexico, each individual whisker is divided into tiny sections and processed in a mass spectrometer.

An island fox that eats the same kind of food (all deer mice or all plant fruit) for a period of time will show an isotope graph that is fairly flat. The mixture of carbon to nitrogen will remain the same. But if an island fox changes up its diet dramatically, a spike will appear in the graph showing a change in the isotopes laid down in the whisker.


Comparing these results for known individual island foxes, living in known habitats, will provide valuable data on how sustained drought impacts island fox diet and how diet impacts their ability to survive and thrive.

Island Fox Whiskers Go to the Lab

FIF Research Grant recipient Juliann Schamel has been collecting island fox whiskers to study the diet of island foxes through stable isotope analysis. Her research project is in conjunction with Dr. Seth Newsome and the Center for Stable Isotopes at the University of New Mexico.


Juliann says "Once I learned how stable isotope ecology works and what we could learn about island fox ecology from this method...I was hooked. The invisible chemical world can reveal fascinating connections within food webs and beyond." More on stable isotopes

"This year, with the support of Friends of the Island Fox and [another conservation fund], we are able to investigate a variety of conservation-geared questions about island fox diet on San Miguel and Santa Rosa Islands. We are looking into how diet differs among habitats on the islands, among demographic groups, and between island fox and island skunk. We are also assessing if and how fox diet has changed over time (2010–present) in response to things like density changes in the fox population and rainfall (drought conditions). This last question may have important implications for the ecology of the acanthocephalan worm, a novel parasite that appeared in the San Miguel Island fox in 2012 (at the beginning of the drought) and has spread through the fox population."

"By using whisker samples that are linked to a known individual with a known history within the island fox monitoring program," Schamel says, "we are able to track the diet of individuals over time to see if and how their diet may have fluctuated during the drought or as they moved to a new habitat."

A number of whisker specimens were collected this winter, including in the beach/dune habitat on Miguel and Rosa where island foxes are not typically counted. This will provide data on dietary resources in the marine adjacent habitat that has not been collected before. Schamel says "I am very excited to run these samples!"


Collected whisker samples are sorted, inventoried, and cataloged. Once they are cleaned, rinsed, and dried, each individual whisker is placed in vial.


Newsome and his lab manager at UNM use a razor and tweezers to divide each whisker into sub-samples: 0.2mg (weighed precisely with a micro-scale).

Schamel explains these sub-samples are fed into "the mass spectrometer, which consumes the samples and spits out data on carbon and nitrogen isotope ratios." The ratios are compared to a library of food resources and their known isotope ratios. "The FIF funding is actually paying the mass spectrometer for its time."

So far ~100 island fox samples from Santa Rosa have been analyzed from a variety of habitats. In addition ~200 island fox whisker samples from San Miguel have been analyzed from 2010–2012, before the drought. Schamel will be at UNM this spring working on samples from 2014–2016. "There are many samples from the same individuals across this time frame, which will be exciting to see!"

Juliann Schamel hopes to present preliminary data at the 2019 Island Fox Conservation Working Group Meeting in May.

Island Fox M152 Has a History

Each island fox counted across the Channel Islands during summer/fall monitoring is a documented individual. They each receive a microchip tag (or PIT tag) like your dog or cat. Why is that important? Why does FIF help fund ID microchips?

When island fox M152 received his radio collar this fall, he already had a documented history.

- He was first captured in 2014, when he was a young adult (estimated 1-2 years old). Island foxes live to be 8-12 years old in the wild. M152 is an adult male in his prime.

- He was living in the same area four years ago, which tells us that he is an adult male with an established territory in this location. Seven pups were seen in this same area in 2018. There is a good possibility that M152 is their father or grandfather. DNA comparison of blood samples could document their relationship. A blood sample was taken from M152.

- 2014 was a drought year and M152 weighed 4.5 lbs (he was considered "thin"). In 2018 he had beefed up to 4.9 lbs (a weight considered "healthy"). A half pound might not seem like much of a difference, but when you are tiny that's almost 10% of your body weight.

- What has made the difference in M152's weight? Is he just eating better this year or is he eating a different diet than in 2014? Four years ago, a whisker sample was taken from M152. This year a whisker sample was collected again. Friends of the Island Fox has funded a Research Grant to evaluate these whisker samples. Whiskers record what the fox has been eating over a four to six month period. Isotopes from the food items, prey or plant matter, are laid down in the whisker as it grows out. M152 may help us learn what an island fox needs to eat to be healthy and successfully parent pups.

FIF Research Grant to Investigate Island Fox Diet Through Whiskers

Island foxes have lots of long black whiskers. These specialized hairs hold important information on diet that could inform decisions on island fox conservation.

Friends of the Island Fox is happy to announce: 

Juliann Schamel is the recipient of the first FIF Research Grant

Schamel is a Biological Science Technician at Channel Islands National Park and the $5,000 grant will help fund research on "The Channel Island Food Web–A Decade of Dietary Resource Use in Channel Island Fox: Implications for Reproduction, Recruitment, and Resilience in a Changing Climate."

It's a big title reflecting the broad range of important data that may be revealed. 

Schamel's work will build on important past research.

In 2009, researchers collected island fox scat samples to look at seasonal diet across all six islands. Their 2014 paper revealed diet varied from island to island and included a higher frequency of insects and fruit than expected. They cautioned that islands with poor native plant diversity offered fewer food options for island foxes in the event of drought.

Just as the science warned, during the consecutive years of the recent drought, island foxes were challenged to find enough food on San Miguel and San Nicolas Islands. Decline of San Nicolas Island fox. During this time a new parasite entered the San Miguel Island fox's diet causing fox deaths.

While scat successfully reveals visual items, (insect exoskeletons, cactus fruit skin and seeds), it may not visually capture everything in the fox's diet. Also, scat data is limited to recording diet in an individual over a short time period, a few days.

In 2010, building on work done on the San Joaquin kit fox, a small sample of island fox whiskers were investigated using stable isotope analysis. 

You are what you eat. Food items (prey and plant) have their own isotope fingerprint. Carbon and nitrogen isotopes consumed by the fox are laid down in the hair shaft documenting an individual's diet over months. 

Since 2010, biologists have been gathering whisker samples from individual island foxes during health checks on the northern islands. These samples have been cataloged by year and with individual fox identification. 

FIF is excited to support Schamel's research using whiskers because it has the potential to:
  • Compare diet between successful mothers and unsuccessful mothers
  • Discover dietary differences between coastal living island foxes and interior living foxes
  • Document changes in diet over the past 9 years, through the drought and as the fox populations have grown
  • Possibly reveal prey items that are a vector for the intestinal parasite on San Miguel Island which has caused fox deaths
Whiskers are amazing! Not only do they offer an opportunity to invest in greater knowledge about the secret lives of island foxes, whiskers grow back and their collection is non-invasive.
